In SCY4 yeast cells, hSOD-33TAG-His6 was co-transformed alongside LeuRS-BH5-T252A in the presence and absence of 1mM 4,5-Dimethoxy-2 nitrobenzylserine, yielding 0.8mg/L mutant hSOD protein with ncAA and none without. Gel electrophoresis and ESI-MS showed high fidelity and confirmed incorporation of 4,5-Dimethoxy-2 nitrobenzylserine.
LeuRS-BH5-T252A was also shown to be effective for the expression of Pho4-GFP fusion constructs in the presence of 4,5-Dimethoxy-2 nitrobenzylserine.
