In DH10B cells, pDule-tfm-Phe1 plasmid bearing tfm-PheRS was co-transformed with pTrc-NTR-124TAG in minimal media, in the presence or absence of 1mM trifluoromethyl-L phenylalanine(tfm-Phe). Full-length protein was produced in the presence of 1mM tfm-Phe only, with yields varying from 12-24mg/L at site 124, and yields varying from 10-16mg/L at site 36.

When incorporating p-Methyl L Phenylalanine targeting site 124, high efficiency and fidelity was observed.
